Abstract

RNA Viruses: A Practical Approach is wide ranging in scope, from emerging technology such as reverse genetics and retrovirus vectors, to money saving tips - how to make your own silica particles for high efficiency RNA extraction and liposomes for cell transfection! Chapter one covers the fundamentals of investigating RNA virus genome structure at a molecular level. Chapters two and three describe techniques for mutagenesis of RNA genomes and analysis of transcription. Chapter four deals with RNA virus-encoded proteinases, an important aspect of the control of RNA virus gene expression. Chapter five considers retrovirus oncogenesis and chapter six analysis of RNA virus quasispecies. Chapter seven describes systems for investigation of in vitro replication of positive-stranded viruses and chapter eight the packaging of RNA virus genomes. In addition to the technical aspects of reverse genetics and retrovirus vectors, both of the final two chapters also consider ethical aspects of these new technologies.

Abstract

This book brings together what is currently known in terms of basic research in the field of long noncoding RNAs (lncRNAs) and builds on this to delve more deeply in the specific roles that lncRNAs are playing during inflammation. Abstract

This book presents a novel method of synthesizing sequence-specific DNA binding pyrrole–imidazole (Py–Im) polyamides and demonstrates polyamides targeting longer base pairs than those previously reported and their cellular application. It discusses the development of the facile synthetic method of tandem hairpin Py–Im polyamides and the method of visualizing telomeres under mild conditions without harsh denaturation, as well as research resulting in the synthesis not only of previously reported tandem dimer polyamides but also longer tandem trimer and tetramer polyamides. Using fluorescently labeled tandem hairpin Py–Im polyamides targeting a human telomere repetitive sequence, the author successfully stained telomeres in fixed cells without denaturation. These fluorescently labeled tandem tetramer polyamides are able to visualize human telomeres with lower background signals than previous polyamide probes, suggesting that they have higher specificity for telomeres. As such, this book provides insights into recent advance in Py–Im polyamides, particularly the extension of the target sites and improvement of the specificity, which can be applied to the study of pyrrole–imidazole polyamides with higher sequence specificity and the function of telomeres.

Abstract

This book describes the development of novel protein-RNA-binding assays and their applications in a high-throughput manner for the identification of small-molecule modulators of protein-RNA interactions to treat cancer and COVID-19. Modulating protein-RNA interactions with small molecules is expected to provide novel biological insights of the interrelation of diseases with the protein-RNA interactome. The modulations may also be exploited therapeutically. For these reasons, the development of a simple, reliable, and sensitive protein-RNA-binding assay is necessary for high-throughput screening to discover new effective chemical entities capable of acting on diverse protein-RNA interactions. This book discusses the discovery of small-molecule modulators targeting protein-RNA interactions that are potentially valuable to treat cancer and COVID-19 by constructing novel high-throughput screening methods. The results of this dissertation provide valuable insights into the regulation of protein-RNA interactions in chemical biology and drug development.
