Sequencing of the human genome and insights into signaling pathways have contributed to the understanding of cancer etiology and the development of new, improved cancer drugs. DNA mutations of a limited set of genes are responsible for the multiple stages of tumorigenesis and metastasis. Matching of therapeutic intervention with insights into the underlying molecular disease mechanism has led to the development of drugs such as Herceptin and Glivec. The deregulation of pathways due to mutated cancer genes provides the conceptual basis for future progress. Will it be possible to extrapolate this principle and derive more efficient drugs targeting cancer pathway components? Potential drug targets have been identified, but our ability to predict the consequences of inhibition of such components is still limited. The state of development of tomorrow’s cancer drugs, directed against growth factors, growth factor receptors and intracellular signaling molecules with kinase activities, is described in this book.

L’Atlante per la stadiazione dei tumori maligni è una pubblicazione ufficiale dell’American Joint Committee on Cancer, riconosciuto leader nella stadiazione delle neoplasie. Questa versione maneggevole, di facile consultazione e riccamente illustrata dell’AJCC Cancer Staging Manual - 6th ed., conferma l’importante ruolo svolto dall’AJCC nella divulgazione dello stato dell’arte della stadiazione TNM, e ne realizza compiutamente lo scopo educativo a vantaggio della comunità medica e degli epidemiologi. L’atlante contiene oltre 400 illustrazioni in bianco e nero appositamente create che descrivono l’estensione anatomica del tumore maligno nella sede primitiva (T), nei linfonodi regionali (N) e nelle metastasi a distanza (M) per varie sedi, tra cui il distretto testa-collo, l’apparato digerente, il torace, l’apparato muscolo-scheletrico, i tessuti molli, la mammella, l’apparato urinario e l’apparato genitale. Ogni illustrazione fornisce precise e minuziose descrizioni atte a chiarire le strutture anatomiche cruciali e a fornire al lettore una visione immediata della progressiva estensione della malattia. Le strutture anatomiche più importanti sono identificate da specifiche definizioni. Tra le caratteristiche più rilevanti del testo vi sono le descrizioni dei criteri diagnostici e di stadiazione più aggiornati riguardo al melanoma, ai tumori ossei e ai carcinomi della mammella, della tiroide, del rene e della prostata. I criteri che hanno motivato la revisione dei principi di stadiazione nei suddetti tumori maligni sono sempre illustrati con l’ausilio di un’iconografia chiara ed efficace. Sono stati inoltre inclusi in ogni capitolo dell’Atlante anche uno schematico "Riassunto delle modifiche" e i paragrafi "Introduzione" e "Anatomia" già presenti nel suddetto AJCC Manual, in modo da poter offrire un utile ripasso ai medici già esperti e un fondamentale testo di consultazione per coloro che si affacciano ora alla professione medica.
