A collection of twenty natural resistant lactic bacteria were isolated from 19 soil and poultry samples coming from Dakar, Senegal. The criteria of selection were based on thermal resistance and resistance to dehydration. A polyphasic characterization, including physiological, biochemical and genetic techniques made it possible to establish a suitable profile for each new isolate. The phenotypical and genetic diversity of the new isolates was demonstrated. Among the most resistant strains, Sp. 20, CWBI-B545 was identified as Lactobacillus paraplantarum ; Sp. 4, CWBI-B534 constitutes a new species of the plantanum group. The species Lactobacillus aminatus sp. nov. was proposed. These two strains present excellent probiotic properties (resistance to acidity, to bile salts and antibiotics). Adding CWBI-B545 in the simulator of the microbial ecosystem of the human gastro-intestinal tract (SHIME) generated beneficial effects on the flora and the production of beneficial metabolites. After freeze-drying, CWBI-B534 and CWBI-B545 are preserved better and longer at room temperature in contrast with control strains. Among the factors responsible for their rusticity, the presence in the cells of a greater fraction of unsaturated fatty acids and the maintenance of the unsaturated fatty acid/saturated fatty acids balance during thermal stress was shown compared to the control strains.
