Abstract

This book provides state-of-the-art reviews of key issues and recent developments relating to chronic myeloid leukemia (CML), acquainting the reader with advances in research, treatment, and promotion of public health. Among the management topics addressed are the choices, advantages, and pitfalls of first-, second-, and third-line treatments; the European LeukemiaNet recommendations; management of adverse effects of tyrosine kinase inhibitors (TKI); management of CML blast crisis; management of pregnancy in the context of CML; the role of hematopoietic cell transplantation; the current experience with TKI discontinuation; and the value of interferon α in improving the outcome of TKI treatment. Various aspects of relevance to treatment outcome are discussed, including prognostic scores, molecular monitoring (principles and interlaboratory standardization), and response-related predictors of survival. Furthermore, updates are provided on the increasing prevalence of CML and its implications and on the changing cost of care for CML, taking into account the forthcoming impact of availability of generic imatinib. .

Abstract

The type I interferon (IFN) signaling pathway is well recognized as a pathway activated by viral infections. It is activated by a variety of microbial pattern recognition receptors including the Toll-like receptors, NOD-like receptors and several cytosolic receptors. Activation of the type I IFN pathway leads to the production of both antiviral factors and products that influence immune cell function. More recently it has been shown that bacteria are also capable of activating this pathway. Bacterial Activation of Type I Interferons reviews both the current understanding of how different bacterial species are able to activate this pathway as well as the influence type I IFNs have on the outcome to infection. Several different bacterial species are covered, spanning Gram positive and Gram negative, intracellular, extracellular, and different host infection sites. An introduction to the pathogenesis of each organism is provided, and the signaling molecules involved in the activation of the type I IFN pathway and the role it plays in animal infection models are also covered.

Abstract

This book introduces readers to Direct Acting Antiviral (DAAs) agents, newly developed drugs to treat chronic hepatitis C virus infection, which have an excellent anti-viral effect on virus replication. These drugs have been developed to enhance the effect of interferon and ribavirin, as the resistant strains against each drug have been described both in vitro and in vivo and the existences of naturally occurring variants have been reported. However, since the combination of different classes of DAAs is sufficient to completely eradicate the virus without the need for interferon or ribavirin, the current treatment regimen does not include interferon. Ribavirin is used in combination with sofosbuvir, a potent polymerase inhibitor, to enhance its antiviral effects. Hepatitis C Virus Treatment offers readers a comprehensive guide to hepatitis C; describing the resistance profiles against these drugs and shedding light on the difficulties involved in DAA therapy and the direction of future treatments, it will equip doctors to understand the essentials of treating the disease. It also describes in detail new and innovative DAA treatments, the effects of the agents, and the characteristics of resistance, providing cutting-edge information not only for hepatologists, but also for researchers, clinical residents, and medical students.

Abstract

Over the past few decades technological advances in transcriptomics, proteomics, metabolomics, and glycomics, along with the ability to selectively knockout genes of interest, have greatly advanced our understanding of the maternal-conceptus interactions that are essential for the establishment and maintenance of a successful pregnancy. This knowledge provides the basis for new research endeavors to address infertility, embryonic loss and recurrent abortion in humans, captive wild animals and important farm species. This book brings together current reviews from leading experts to address the diversity of mechanisms by which species establish and maintain pregnancy. Implantation in rodents, dogs, pigs, cattle, sheep, horses, primates, humans and embryonic diapause in wild species are discussed. The reviews provide readers with the latest knowledge on the role of the endometrial steroid receptors, adhesion factors, cytokines, interferons, steroids, prostaglandins, growth factors and immune cells involved in the regulation of conceptus development. .
