Abstract

Single Subject Designs in Biomedicine draws upon the rich history of single case research within the educational and behavioral research settings and extends the application to the field of biomedicine. Biomedical illustrations are used to demonstrate the processes of designing, implementing, and evaluating a single subject design. Strengths and limitations of various methodologies are presented, along with specific clinical areas of application in which these applications would be appropriate. Abstract

(Draft) In this volume the unique and common features of rhabdoviruses, particularly their morphological, molecular, and pathogenic characteristics, and their phylogenetic relationships are reviewed. Rhabdoviruses have a very wide host range and have been isolated from plants, insects, and almost from all vertebrates including fish and primates. The Rhabdoviridae family consists of six genera which have all been associated with diseases. While rhabdoviruses that are etiological agents of human diseases can cause serious public health problems, other members of this family that infect domestic livestock and agricultural plants can also cause enormous economic loss. .

Abstract

The articles in this volume have been selected to demonstrate the progress in the development of human antibody therapeutics for viral disease. Keck et al. review the nature of the immune response to the Hepatitis C virus (HCV) and the details of viral neutralization by antibodies, providing a conceptual model for the clinical use of HCV-specific antibodies. Huber et al. summarize the initial clinical experiences with antibody therapeutics for Human Immunodeficiency Virus that can be targeted to either the HIV virion or to host cell proteins. A discussion of the breadth immune strategies that is required to control human rabies is provided by Nagarajan et al., with a particular focus on India and other countries in which rabies is endemic. The development of pavilizumab for RSV prophylaxis is reviewed in Wu et al., in addition to results of antibody optimization studies that provide surprising insights and have broad general implications for anti-viral antibody engineering. Melhop and Diamond explicate the biology of West Nile Virus as a general model for flaviviruses, while using their cloned antibodies as a springboard to consider the mechanisms of WNV neutralization. The volume concludes with a description of methods to clone human antibodies in their native configurations, which access a class of antibodies that differ from those obtained by recombinant DNA or transgenic mouse methods. The articles in this volume are definitive and comprehensive reviews written by experts who have sought to define the principles of viral neutralization by human antibodies. Abstract

Because of its highly contagious nature research work on FMDV is restricted to a small number of laboratories worldwide that have adequate containment facilities. Abstract

Pathogenic bacteria for human and animals have developed sophisticated weapons, termed virulence factors, to ensure their replication and persistence into their hosts. The first role of these virulence factors is to loosen the host defenses against microorganisms represented by innate and adaptive immunities. Small GTPases of the Rho subfamily have been shown top play important functions in theses systems. Abstract

Reoviridae family members are large, complex viruses that infect human, animals, plants and insects. They are unique in that they lack lipid envelopes and package their genomes of discrete double-stranded segments of RNA within multi-layered capsids. Lack of a lipid envelope has allowed three-dimensional structures of these large complex viruses to be obtained. The capsids of these viruses undergo cell entry, uncoating, the enzymatic functions necessary for transcription of the genome, and are later involved in egress from host cells. Recent years have seen an increase in our knowledge of the structure of these viruses coupled with substantial progress in unravelling the molecular details of these processes. Intriguingly, despite their diversity in hosts, structures and modes of transmission, striking parallels have emerged in the molecular interactions necessary for the essential processes of virus entry, assembly and release. This book reviews our current understanding of Reoviridae entry, disassembly/assembly and egress in addition to updating high resolution structures of viral proteins and capsids from three different genera of the family.

Abstract

The earliest observation of cytomegalovirus (CMV) interactions with the host cell was owl eye cytopathology in various tissues. It was recognized in the early 1970s that human CMV caused in utero infections resulting in congenital brain damage and other sensory neurological complications. Events of the 1980s and early 1990s, such as the wide application of solid organ and bone marrow transplantation and the emergence of AIDS, put the spotlight on human CMV. We understood that the virus was an opportunistic agent associated with immunosuppression. The golden age of cytomegalovirus research was ushered in during the late 1970s and early 1980s by a set of powerful new technologies that included restriction enzymes, DNA cloning, DNA sequencing, and open reading frame prediction. The genetic manipulation and propagation of novel CMV strains was accelerated with the app- cation of bacterial artificial chromosome technology. Today, we still struggle to understand the full spectrum of disease associated with human CMV. To the molecular biologist, CMV is a master of regulation in the eukaryotic cell where it either replicates or remains latent. To the immunologist, CMV is a master of immune evasion with tools to escape both the innate and acquired immune responses. The use of animal models with non-human CMVs has become significantly more sophisticated and tied to a more certain understanding of the interrelationships of non-human and human CMV genes.
