There are no shortcuts in organic chemistry. Understanding and mastery cannot be achieved without devoting adequate time and attention to the theories and concepts of the discipline. It is with this premise that Ouellette and Rawn set out to comprehensively survey the field in Organic Chemistry, with an emphasis on connecting the basic principles of organic chemistry to real world challenges that require analysis, not just recall. Organic chemistry is at the interface of physical and biological science, and this new text lays out the basic principles of organic chemistry in their relation to a host of other fields in both physical and biological sciences. Chemical bonding determines molecular structure, which in turn dictates physical, chemical, and biological properties from the smallest molecules to the largest. Molecular structure determines reaction mechanisms, from the smallest to the largest molecules. Reaction mechanisms determine strategies for organic synthesis, and these synthetic principles extend to every aspect of synthesis, from drug design to the methods cells employ to synthesize the molecules of which they are made. These relationships form a continuous narrative throughout the book, wherein principles logically evolve from one to the next, from the simplest to the most complex examples, with abundant connections between the text and real world applications. Contains extensive examples of biological relevance Includes an important chapter on organometallic chemistry not found in other standard references Extended, illustrated glossary Appendices on thermodynamics, kinetics, and transition state theory.

This volume on iron-sulfur proteins includes chapters that describe the initial discovery of iron-sulfur proteins in the 1960's to elucidation of the roles of iron sulfur clusters as prosthetic groups of enzymes, such as the citric acid cycle enzyme, aconitase, and numerous other proteins, ranging from nitrogenase to DNA repair proteins. The capacity of iron sulfur clusters to accept and delocalize single electrons is explained by basic chemical principles, which illustrate why iron sulfur proteins are uniquely suitable for electron transport and other activities. Techniques used for detection and stabilization of iron-sulfur clusters, including EPR and Mossbauer spectroscopies, are discussed because they are important for characterizing unrecognized and elusive iron sulfur proteins. Recent insights into how nitrogenase works have arisen from multiple advances, described here, including studies of high-resolution crystal structures. Numerous chapters discuss how microbes, plants, and animals synthesize these complex prosthetic groups, and why it is important to understand the chemistry and biogenesis of iron sulfur proteins. In addition to their vital importance in mitochondrial respiration, numerous iron sulfur proteins are important in maintenance of DNA integrity. Multiple rare human diseases with different clinical presentations are caused by mutations of genes in the iron sulfur cluster biogenesis pathway. Understanding iron sulfur proteins is important for understanding a rapidly expanding group of metabolic pathways important in all kingdoms of life, and for understanding processes ranging from nitrogen fixation to human disease.

Metallothioneins (MTs) represent a large family of proteins characterized by high heavy metal content (mainly CuII and ZnII) and by an unusual cysteine abundance. They have a powerful protective function in all animal tissues, due most likely to their properties as free radical scavengers protecting against oxidative damage. Moreover, the presence and overexpression of MTs in various pathological conditions, such as metal dyshomeostasis, cell proliferation, neurological disorders, and chemotherapy and radiotherapy resistance, could be used as an important prognostic marker, as a histopathologi
