Abstract

Respiratory syncytial virus (RSV) causes seasonal epidemics during the winter and wet seasons, particularly in young children. Although there is one drug available for prophylaxis, palivizumab, it is indicated for high-risk children only. Therefore a vaccine is urgently needed. This book provides an overview of RSV and discusses its incidence and presentation in different populations, including preterm infants and children with congenital heart disease. It also discusses future strategies for RSV prophylaxis and developments in and barriers to creating a viable vaccine.

Abstract

RNA Viruses: A Practical Approach is wide ranging in scope, from emerging technology such as reverse genetics and retrovirus vectors, to money saving tips - how to make your own silica particles for high efficiency RNA extraction and liposomes for cell transfection! Chapter one covers the fundamentals of investigating RNA virus genome structure at a molecular level. Chapters two and three describe techniques for mutagenesis of RNA genomes and analysis of transcription. Chapter four deals with RNA virus-encoded proteinases, an important aspect of the control of RNA virus gene expression. Chapter five considers retrovirus oncogenesis and chapter six analysis of RNA virus quasispecies. Chapter seven describes systems for investigation of in vitro replication of positive-stranded viruses and chapter eight the packaging of RNA virus genomes. In addition to the technical aspects of reverse genetics and retrovirus vectors, both of the final two chapters also consider ethical aspects of these new technologies.

In the spring of 2013, the influenza A (H7N9) virus shocked China, and caused 134 human infections with 45 deaths in its first influenza season. In the 2013-2014 winter, the virus struck China again and caused 273 cases with 108 deaths. This H7N9 outbreak has raised concern for another influenza pandemic. In this book, the epidemiology of the H7N9 outbreak, and the origin and diversity of the virus genome are discussed. Abstract

Annotation Recombinant viruses expressing reporter fluorescent or bioluminescent proteins are an excellent option to evaluate the dynamics of viral infection progression in both cultured cells and/or validated animal models of viral infection. Reporter proteins are valid surrogates for direct detection of infected cells in vitro and in vivo, without the use of secondary methodologies to identify infected cells. By eliminating the need of secondary labeling, tractable replicating-competent, reporter-expressing viruses provide an ideal approach to monitor viral infections in real time, representing a significant advance in the study of the biology of viruses, to evaluate vaccination approaches, and to identify new therapeutics against viral infections using high-throughput screening settings. In this Special Issue "Replication-Competent Reporter-Expressing Viruses" we review replication-competent, reporter-expressing viruses belonging to different families, methods of characterization, and applications to facilitate the study of in vitro and in vivo viral infections. We also seek to discuss disadvantages and limitations associated with these reporter-expressing viruses. Finally, we provide rational future perspectives and additional avenues for the development, characterization, and applications of recombinant, reporter-expressing, competent viruses.
