Abstract

There is considerable interest today for the reactions of anticancer metallodrugs with proteins as these interactions might feature processes that are crucial for the bio-distribution, the toxicity and even the mechanism of action of this important group of anticancer agents. Valuable structural and functional information on these adducts could be derived from several biophysical studies mainly relying on the application of X-ray diffraction and ESI MS techniques. The structural and functional information achieved on the respective metallodrug–protein adducts allowed us to identify some general trends in the reactivity of anticancer metallodrugs with protein targets.

Abstract

The present work makes use of coarse-grained molecular dynamics simulations and continuum models to investigate the behaviour of biomolecules in experiments such as mechanical pulling and driven transport across nanopores. The approach reproduces the wide phenomenology of experiments and allows one to maintain the main features of the transport by modelling the process as a 1D dynamics in a suited potential of the mean force. The standard 1D continuum view is enriched by proposing a model for the description of the shape of isolated molecules based on a tensorial representation and Cauchy-Born rule. Results indicate limitations for unconstrained dynamics and appropriateness for driven ones.

Abstract

Few would have predicted 20 years ago that myosins constitute a superfamily with at least two-dozen classes and that these molecular motors are involved in a multitude of intracellular activities including cell division, cell movement, intracellular transport and signal transduction. Application of state-of-the-art cellular and molecular biological, structural biological, genetic, biochemical and biophysical techniques has provided and continues to provide critical information regarding the structure–function relationship; and the cellular roles of various myosins in organisms as diverse as protozoa, yeast, plants and higher animals. The association of myosins with diseases including neurological disorders, immu- deficiencies, cardiomyopathies, hearing and vision loss testify to the importance of understanding the biochemical properties and cellular roles of myosins. The 16 chapters in this volume summarize the tremendous progress made in studying members of the myosin superfamily in recent years and offer critical insight into what future research will yield. I would like to express my sincere gratitude to the authors of this volume. It was a pleasure to work with each of you and I thank you for the considerable efforts in making this international endeavor possible. I also thank John Trinick from the University of Leeds, UK, for the elegant montage of images of single molecules of myosins on the cover, which beautifully shows the structures of some of these amazing molecules. Abstract

Cet ouvrage couvre les différents aspects de la structure des protéines : leur composition chimique, l’établissement de leur structure, les différents niveaux de leur organisation, les aspects énergétiques de leur stabilité, leur dynamique moléculaire et la prédiction théorique de leur structure, prenant en compte les résultats substantiels et significatifs obtenus au cours des dernières années. Le premier chapitre concerne la chimie de leurs constituants de base, les acides aminés, connaissance qui est précieuse pour les chimistes des protéines et les enzymologistes mais dont les données sont dispersées dans la littérature. Pour la première fois dans ce type d’ouvrage un chapitre entier est consacré aux protéines membranaires, dans la connaissance desquelles des progrès considérables ont été faits au cours des dernières années. Suit une description des mécanismes très élaborés de l’insertion de ces protéines dans les membranes biologiques. Un chapitre est consacré à la dynamique conformationnelle de ces macromolécules, qui joue un rôle fondamental dans leurs propriétés biologiques. Les logiciels qui permettent de prédire la structure des protéines sur la base de leur composition en acides aminés sont également présentés. D’un point de vue pédagogique, certains chapitres comportent un exposé plus ou moins bref de l’histoire de certaines découvertes importantes ou la logique d’évolution de concepts majeurs ou de techniques expérimentales de haut niveau.
