Abstract

With cardiometabolic diseases still topping the list of mortality causes and in facing the obesity and diabetes epidemic, there remains a great need to better understand the pathophysiological derangements underlying these conditions. During the past years, it has become increasingly appreciated that low grade systemic inflammation is a common hallmark of cardiometabolic disorders--not only concerning diabetes and atherosclerotic cardiovascular disease but also involving non-alcoholic fatty liver disease. Recently developed high-throughput laboratory techniques for lipidomics and metabolomics have enabled researchers to discern novel crosstalk pathways between lipid phenotypes and enhanced chronic inflammation. With this Special Issue of the Journal of Clinical Medicine, entitled "Novel Aspects of Lipoprotein Metabolism with a Focus on Systemic Inflammation", researchers were invited to submit original papers and reviews on various topics, in particular, at the interface of lipid metabolism and inflammation.

Abstract

Dyslipidemia is a major risk factor for cardiovascular events, cardiovascular mortality, and all-cause mortality. The earlier in life dyslipidemia is treated, the better the prognosis. The current book is an excellent one on dyslipidemia written by experts on this topic. This book includes 12 chapters including 5 on lipids, 4 on hypercholesterolemia in children, and 3 on the treatment of dyslipidemia. This book should be read by all health care professionals taking care of patients, including pediatricians since atherosclerotic cardiovascular disease begins in childhood.

Abstract

Abstract

The purpose of this issue is to present the impact in clinical practice as well as in medical research of novel molecules that have been introduced in the treatment of diabetes mellitus, dyslipidemia, and cardiovascular disease. The topic focuses on the improvements in disease outcomes as well as the numerous beneficial effects of use of certain medications such as GLP-1 agonists, dual GLP-1/ GIP agonists, and SGLT-2 inhibitors for diabetes mellitus treatment; PCSK9 inhibitors and small interfering RNA (siRNA) molecules for dyslipidemia; and antiaggregants, oral anticoagulants, scubitril/valsartan, and non-steroidal mineralocorticoid receptor agonists or dapagliflozin in cardiovascular diseases. We believe that the number of real-life studies and in vivo and in vitro research of the effects of these drugs is quite low at the moment, leading to a lack of understanding of the complex molecular effects and pleiotropic effects of these medications, as well as incomplete knowledge regarding the repurposing of these drugs in clinical practice. Thus, this Special Issue is incredibly topical and important, and we look forward to your submissions which will help to clarify the issues surrounding such drugs.

With the Karlsruhe Heart Model (KaHMo) we aim to share our vision of integrated computational simulation across multiple disciplines of cardiovascular research, and emphasis yet again the importance of Modelling the Human Cardiac Fluid Mechanics within the framework of the international STICH study. The focus of this work is on integrated cardiovascular fluid mechanics, and the potential benefits to future cardiovascular research and the wider bio-medical community.
