Chromosomes, as the genetic vehicles, provide the basic material for a large proportion of genetic investigations, from the construction of gene maps and models of chromosome organization, to the inves tigation of gene function and dysfunction. The study of chromosomes has developed in parallel with other aspects of molecular genetics, beginning with the first preparations of chromosomes from animal cells, through the development of banding techniques, which permitted the unequivocal identification of each chromosome in a karyotype, to the present analytical methods of molecular cytogenetics. Although some of these techniques have been in use for many years, and can be learned relatively easily, most published scientific reports—as a result of pressure on space from editors, and the response to that pressure by authors—contain little in the way of technical detail, and thus are rarely adequate for a researcher hoping to find all the necessary information to embark on a method from scratch. A new user needs not only a detailed description of the methods, but also some help with problem solving and sorting out the difficulties en countered in handling any biological system. This was the require ment to which the series Methods in Molecular Biology is addressed, and Chromosome Analysis Protocols forms a part of this series.

Er is binnen de medische wetenschappen geen vakgebied waarin momenteel zo opvallend veel vooruitgang wordt geboekt als in de genetica. Dit gegeven vormde de belangrijkste overweging om reeds na vijf jaar Medische genetica grondig te herzien. Ook deze nieuwe druk is weer bedoeld als basisstof voor de opleiding van studenten in de geneeskunde en de nascholing van huisartsen en specialisten. Daarnaast echter is de stof uitermate bruikbaar voor andere disciplines, zoals de medische biologie. Bij de opzet van deze nieuwe druk is de indeling van de hoofdstukken enigszins aangepast. Elk hoofdstuk wordt voorafgegaan door een korte karakteristiek en waar mogelijk afgesloten met een samenvatting. Aan de meeste hoofdstukken zijn bovendien enkele toetsvragen toegevoegd (de antwoorden zijn achterin opgenomen). De opbouw van de tekst is erop gericht de basisarts op zo toegankelijk mogelijke manier de kennis te verschaffen die nodig is om het mogelijk erfelijke karakter van een ziektebeeld te onderkennen en iets te begrijpen van de psychosociale problemen die een erfelijke aandoening met zich meebrengt. Op deze wijze is hij in staat ouders over erfelijkheid voor te lichten en te begeleiden. Regelmatig zullen deze moeten worden doorverwezen naar een klinisch- genetisch centrum waar met de specialisten zal moeten worden overlegd. De inhoud van het boek is in grote lijnen hetzelfde gebleven. In een inleidend hoofdstuk wordt een overzicht gegeven van de belangrijkste nieuwe ontwikkelingen en de betekenis hiervan voor de erfelijkheidsadvisering. Tegelijkertijd wordt duidelijk dat de medisch- technologische vooruitgang vragen oproept van juridische, psychosociale en ethische aard. De evolutie is niet meer als afzonderlijk hoofdstuk opgenomen maar wordt behandeld in het hoofdstuk over populatiegenetica. Er zijn twee nieuwe hoofdstukken: een over aangeboren afwijkingen en een over de maatschappelijke en ethische consequenties van erfelijkheidsonderzoek. De hoofdstukken over recombinant- DNA-technologie, overervingspatronen, metabole stofwisselingsstoornissen, multifactoriële overerving, genetica van kanker, populatiegentica en mentale retardatie werden geheel herschreven

Genes V gives an integrated account of the structure and function of genes in both prokaryotic and eukaryotic organisms. It is thoroughly up-to-date with the latest thinking and research in the field. Successive editions have provided an integrated account of the whole field of modern molecular genetics and this edition continues that approach, providing a new synthesis with a much greater emphasis on how genes function in their biological context. The book begins with an introductory overview of material that the student should have encountered in previous courses. These first two chapters consider the cell as an assembly of macromolecules, reviewing the function of these molecules and how they are organized into cell organelles. There follows a detailed examination of the role and function of DNA with increased emphasis on genetic mapping in the context of human disease. The third part of the book discusses translation and has been thoroughly updated to reflect new knowledge about structure and the specificity on DNA function. A new section on cell biology follows. This brings together much information that was scattered in previous editions of the book and integrates it with new knowledge on protein transport, signal transduction, and the cell cycle. The section on gene expression has been thoroughly modernized and updated as have the two chapters which follow, one of which gives an entirely new account of gene sizes, organization and differences in eukaryotes. The book gives, for the first time, a unifying concept for eukaryotic transcription and its regulation reflecting work done since 1990. The chapter on recombination has been entirely rewritten and is followed by updated coverage of manipulations of DNA: transposons, retroviruses, and rearrangements.
