Abstract

Many recent advances in experimental instrumentation, reagents, and imaging technology have dramatically expanded the range of tools available for the study of the cytoskeleton. In Cytoskeleton Methods and Protocols, Ray Gavin brings together an international panel of experienced researchers to detail the readily reproducible methods that utilize biochemistry, immunology, genetics, microscopy, and image analysis for investigating cytoskeleton structure and function. Each protocol contains proven step-by-step instructions that enable both the novice and experienced researcher to achieve successful experimental results. The protocols utilize diverse model systems in a variety of organisms, including Saccharomyces, Micrasterias, Tetrahymena, Drosophila, Spisula, and Xenopus. Microscopy applications include digital-video microscopy and computer-assisted systems for the evaluation of cell motility and morphology. Help notes and tips accompany each protocol and provide additional, often unpublished, information that can make the difference between success and failure. State-of-the-art and highly practical, Cytoskeleton Methods and Protocols makes available a diverse collection of powerful experimental systems and tools for successfully studying cytoskeleton structure and function.

Abstract

Nonviral gene therapy-an emerging field with great clinical promise-avoids many problems associated with viral gene therapy and offers the possibility of superior clinical effectiveness. In Nonviral Vectors for Gene Therapy: Methods and Protocols, Mark A. Findeis has assembled a panel of active researchers to present their best methods not only for preparing, handling, and characterizing gene delivery agents, but also for gene delivery. To help those preparing and characterizing gene transfer agents, the contributors examine a broad range of compounds that bind with DNA to form the compact complexes that facilitate cellular delivery-among them peptide conjugates, synthetic polymers, and lipids. They also outline specific approaches to gene transfer in vivo, including direct delivery by intratumoral injection and indirect delivery by cell-specific targeting of DNA complexes, and discuss in detail many spectroscopic techniques for characterizing nonviral gene delivery agents. Opportunities for the development and application of these novel vectors in the research lab and eventually, in the clinic, are highlighted. Comprehensive and state-of-the-art, Nonviral Vectors for Gene Therapy: Methods and Protocols illuminates for today's investigators the powerful new approaches to the creation of nonviral materials, as well as their efficacious use in delivering therapeutic genes to patients across the spectrum of human disease.

Abstract

It is now clear that kinesin-like-proteins (KLP), generally thought to be responsible for the transport of cellular cargoes, are involved in many different cellular processes now being widely investigated. In Kinesin Protocols, Isabelle Vernos and a panel of hands-on experts present their most productive and reproducible techniques for the identification, purification, and characterization of the kinesin superfamily of microtubule-dependent motors. The methods range from the most basic to the most sophisticated and include step-by-step instructions and extensive cautionary notes to ensure experimental success. Among the approaches discussed are methods to express and purify kinesins in different systems, to characterize microtubule-enhanced ATPase activity and motility properties, and to test microtubule destabilizing activity. Detailed examples of how to address functional studies are also presented, along with some very new methods for studying the role of KLP in the organization of microtubules in three dimensions. There are also advanced methods for the study of kinesins at the structural level. Comprehensive and highly practical, Kinesin Protocols makes available all the key basic and cutting-edge methods needed successfully to study the multifaceted world of kinesin-like proteins and to explore their many functions.

Abstract

Adipose tissue is now recognized as a widely dispersed secretory organ that exhibits autocrine, paracrine, and endocrine properties, and plays a significant role in obesity, the most common health problem in industrialized countries. In Adipose Tissue Protocols, Gérard Ailhaud and a team of laboratory experts and clinicians describe in step-by-step detail the major techniques needed for the study of adipose tissue and cells. Drawn from both in vivo and in vitro studies, these readily reproducible methods cover a broad range of techniques, including the choice of adipose tissue depot and of morphological techniques for work on both brown adipose tissue (BAT) and white adipose tissue (WAT). Major treatment is accorded the isolation, subcellular fractionation, and transfection of low density adipocytes, as well as the metabolic aspects of nutrient uptake and key assays of nutrient and ion fluxes. Also covered are: biopsies and quantification of lipid-related mRNAs; cultures of adipose precursor cells from WAT and BAT; measurements of adipose secretory products; assessment of WAT metabolism in vivo; and assays of lipid-related enzymes. Innovative and highly practical, Adipose Tissue Protocols offers endocrinologists, physiologists, cell biologists, and pharmacologists a gold-standard collection of proven methods for effective nutritional, physiological, and molecular-level research on adipose tissue.

Abstract

An essential element in direct cell-cell communication between virtually all cell types in vertebrates, connexins have been recently linked to an increasing number of human genetic diseases. In Connexin Methods and Protocols, well established investigators describe in detail their best techniques for studying the cellular and molecular biology of connexins, as well as their physiological properties. These forefront methods are specifically designed to investigate the life of connexins from biosynthesis to assembly into gap junction channels, and also include powerful assays to perform functional studies on these routes of cell-cell communication. To ensure experimental success, each time-tested technique contains detailed step-by-step instructions, as well as tips on how to avoid pitfalls. Also discussed are the many specific theoretical aspects of connexin biology and physiology. Cutting-edge and highly practical, Connexin Methods and Protocols offers laboratory researchers a comprehensive collection of proven methods for studying the role of connexins in intercellular communication and human genetic disorders, as well as finding promising new therapeutics for those disorders.

Abstract

Nucleases, the enzymes that cleave and shape nucleic acids, are key regulators of cellular metabolism. Angiogenesis and the unfolded protein response are just a few instances of important intra- and extracellular pathways regulated by nucleases. In Nuclease Methods and Protocols, researchers at the forefront of academic research and pharmaceutical drug development from four continents summarize their recent results. Their projects involve nucleases in many different areas of medicine and biology, ranging from protein structure and folding, to DNA repair, to developing cures for insidious diseases such as cystic fibrosis, AIDS, and cancer. Each chapter contains a minireview of a specific nuclease or nuclease-related theme, a discussion of why and when to use the assays described, and often, an explanation of how the assay was developed. The authors describe their most valuable methods for determining the mode of action, structure, interaction with other molecules, and physiological role of nucleases. Detailed step-by-step instructions and notes on avoiding pitfalls help ensure readily reproducible results. Comprehensive and highly practical, Nuclease Methods and Protocols offers both novice and experienced researchers a deeper understanding of the importance of nucleases in cell metabolism, growth control mechanisms, and, increasingly, in diagnostics and therapeutics.
