Abstract

Prodrugs are substances administered in an inactive form that is then metabolized in the body in vivo into the active compound. The rationale behind administering prodrugs is to optimize absorption, distribution, metabolism, and excretion of these drugs. Since first described in the 1950s, prodrugs continue to be a fertile area of research. There are a number of small pharmaceutical/biotech companies dedicated to using prodrugs for the delivery of older but problematic drugs as well as to developing broad-based prodrug technologies for application to new and future drugs. These volumes represent a comprehensive guide to prodrugs and will guide the reader through the current status of the prodrug concept and its many applications and to highlight its many successes in overcoming formulation and delivery of problematic drugs.

Abstract

Il processo di identificazione è per una droga vegetale di enorme importanza ai fini dell'efficacia e sicurezza della stessa. Infatti la risposta terapeutica e gli effetti indesiderati sono fortemente condizionati dall'identificazione della droga e dalla presenza di contaminanti botanici. Il testo si compone di due parti; la prima sintetizza gli aspetti diagnostici che indirizzano verso il riconoscimento della droga vegetale, mentre la seconda descrive gli aspetti macro- e microscopici della droga ed i saggi chimici e farmacologici che consentono con certezza l'identificazione della stessa. Infine, l'ultima parte riporta alcuni esempi di cromatogrammi e di tracciati di organi isolati. Il volume sarà utile agli studenti e a tutti coloro che, per la loro attività professionale, devono identificare le droghe vegetali ed evidenziare eventuali corpi vegetali estranei.

Abstract

In 1860, Oliver Wendell Holmes pointedly expressed himself to the Massachusetts Medical Society: I firmly believe that if the whole Material Medica, as now used, could be sunk to the bottom of the sea, it would be all the better for mankind, and all the worst for the fishes.  Should one think the same about the current approach in drug discovery from plants? Probably yes. Despite the spending of billions of US dollars, and three decades of efforts, high-throughput screenings have only allowed the discovery of a couple of drugs. One could have reasonably expected the discovery of an arsenal of drugs from the millions of plant extracts randomly tested, but hits  can be inactive in vitro or too toxic, some molecules need to be metabolized first to be active, and false-positive and false-negative results are common. The bitter truth is that the robotic approach in discovering drugs from plants has proven, to date, its inability to excavate the hundreds of molecules that will contribute to the health progress of Man. However, one can reasonably see that the last patches of primary rainforest on earth hold still hundreds of spectacularly active drugs that await discovery.
