Abstract

In this third edition of his popular undergraduate-level textbook, Des Nicholl recognises that a sound grasp of basic principles is vital in any introduction to genetic engineering. Therefore, the book retains its focus on the fundamental principles used in gene manipulation. It is divided into three sections: Part I provides an introduction to the relevant basic molecular biology; Part II, the methods used to manipulate genes; and Part III, applications of the technology. There is a new chapter devoted to the emerging importance of bioinformatics as a distinct discipline. Other additional features include text boxes, which highlight important aspects of topics discussed, and chapter summaries, which include aims and learning outcomes. These, along with key word listings, concept maps and a glossary, will enable students to tailor their study to suit their own learning styles and ultimately gain a firm grasp of a subject that students traditionally find difficult.

Abstract

Horizontal gene transfer is a major driving force in the evolution of many bacterial pathogens. The development of high-throughput sequencing tools and more sophisticated genomic and proteomic techniques in recent years has resulted in a better understanding of this phenomenon. Written by leading experts in the field, this edited volume is aimed at graduate students and researchers and provides an overview of current knowledge relating to the evolution of microbial pathogenicity. This volume provides an overview of the mechanisms and biological consequences of the genome rearrangements resulting from horizontal gene transfer, in both prokaryotes and eukaryotes, as well as overviews of the key mobile genetic elements involved. Subsequent chapters focus on paradigms for the evolution of important bacterial pathogens, including Salmonella enterica, Streptococcus pneumoniae, and Staphylococcus aureus. The influence of socioeconomic parameters in the dissemination of transferable elements, such as antibiotic resistant genes in bacteria, is also discussed.

Abstract

Once per life cycle, mitotic nuclear divisions are replaced by meiosis I and II – reducing chromosome number from the diploid level to a haploid genome, reshuffling the homologous chromosomes by their centromeres, and recombining chromosome arms by crossing-over. In animals, including humans, all this happens during the germ cell formation of eggs and sperm. Due to the reign of meiosis, no child is a true genetic copy of either parent. Central to mainstream meiosis, the mechanisms of reciprocal exchange at crossover/chiasma sites stand out as a controlled program of biologically significant molecular changes. To initiate the meiotic exchange of DNA, surgical cuts are made as a form of calculated damage that is subsequently repaired by homologous recombination. These key events are accompanied by ancillary provisions at the level of chromosome core organization, sister chromatid cohesion, and differential centromere connectivity. Great progress has been made in recent years to further our understanding of these mechanisms. Questions still open primarily concern the placement of and mutual coordination between neighboring crossover events. The current book addresses these processes and mechanisms in multicellular eukaryotes, such as Drosophila, Arabidopsis, mice and humans. The pioneering model systems of yeasts, as well as evolutionary aspects, will be addressed in a forthcoming volume.

Abstract

Like the geological treasures that surely must lie below the Athabasca glacier, some of the answers to key questions about the control of respiration remain buried. But bit by bit, year by year the ice is melting and perhaps in some areas the rocks are starting to be revealed. With a mission to advance our understanding of the emerging th th gems of respiration, the X Oxford Conference was held between 19–24 September (2006) next to the turquoise and tranquil waters of Lake Louise, in the middle of Banff National Park, Alberta, Canada. Since its inauguration 30 years ago in Oxford, the Oxford Conference on mo- ling and control of breathing has been held every three years in locations spanning the globe (a list of past conferences and publications that have emerged is included in the pages that follow). The series has provided key opportunities for respiratory scientists to meet with colleagues, discuss recent advances and celebrate their field. The 2006 Lake Louise meeting was Canada’s second Oxford Conference; the th previous Canadian meeting (the VII Oxford Conference) was held in 1997 at the Grandview Inn in Huntsville (Ontario) and was chaired by Richard Hughson, David Cunningham and Jim Duffin.

Abstract

Parasitic, bacterial and viral agents continue to challenge the welfare of humans, livestock, wild life and plants worldwide. The public health impact and financial consequences of these diseases are particularly hard on the already overburdened economies of developing countries especially in the tropics. Many of these disease agents utilize insect hosts (vectors) to achieve their transmission to mammals. In the past, these diseases were largely controlled by insecticide-based vector reduction strategies. Now, many of these diseases have reemerged in the tropics, recolonizing their previous range, and expanding into new territories previously not considered to be endemic. Habitat change, irrigation practices, atmospheric and climate change, insecticide and drug resistance as well as increases in global tourism, human traffic and commercial activities, have driven the reemergence and spread of vector borne diseases. While these diseases can be controlled through interventions aimed at both their vertebrate and invertebrate hosts, no effective vaccines exist, and only limited therapeutic prospects are available for their control in mammalian hosts. Molecular technologies such as transgenesis, which is the subject of this book, stand to increase the toolbox and benefit disease management strategies.

Abstract

Once per life cycle, mitotic nuclear divisions are replaced by meiosis I and II—reducing chromosome number from the diploid level to a haploid genome and recombining chromosome arms by crossing-over. In animals, all this happens during formation of eggs and sperm—in yeasts before spore formation. The mechanisms of reciprocal exchange at crossover/chiasma sites are central to mainstream meiosis. To initiate the meiotic exchange of DNA, surgical cuts are made as a form of calculated damage that subsequently is repaired by homologous recombination. These key events are accompanied by ancillary provisions at the level of chromatin organization, sister chromatid cohesion and differential centromere connectivity. Great progress has been made in recent years in our understanding of these mechanisms. Questions still open primarily concern the placement of and mutual coordination between neighboring crossover events. Of overlapping significance, this book features two comprehensive treatises of enzymes involved in meiotic recombination, as well as the historical conceptualization of meiotic phenomena from genetical experiments. More specifically, these mechanisms are addressed in yeasts as unicellular model eukaryotes. Furthermore, evolutionary subjects related to meiosis are treated.

Abstract

GENETIC DEMOCRACY involves an in-depth analysis of the ethical, social and philosophical issues related to modern genetic research and gene technology. The aim of the book is to introduce systematic research on the social and ethical impacts of the use and development of genetically modified organisms (GMOs) as well as the acquisition, use and storage of human genetic information (HGI). The book contributes into enhancing public discussion and reaching fair and democratic decision-making practices in GMO and HGI use and development both on local and global level. There are currently few European texts which address the issues involved in a theoretical and systematical manner. GENETIC DEMOCRACY have been written from the viewpoint of social and political philosophy rather than that of traditional bioethics. There is a clear need for a throughout and authoritative philosophical and ethical analysis of the issues involved in genetic research and gene technology. The book will appeal to philosophers, social scientists, genetics professionals, policy makers, academics, industrial organisations and human rights organisations as well as university students and legal scholars. The book will have a broad appeal across Europe, Asia and America since many states are currently considering policy responses to many of the practices discussed in the books (e.g., human biobanks).

Abstract

Mapping of animal genomes has generated huge databases and several new concepts and strategies, which are useful to elucidate origin, evolution and phylogeny. Genetic and physical maps of genomes further provide precise details on chromosomal location, function, expression and regulation of academically and economically important genes. The series Genome Mapping and Genomics in Animals provides comprehensive and up-to-date reviews on genomic research on a large variety of selected animal systems, contributed by leading scientists from around the world. This volume summarizes the first era of genomic studies of aquaculture species, in which the tools and resources necessary to support whole-genome sequencing were developed. These tools will enhance efforts toward selective breeding of aquaculture species. Included in this volume are summaries of work on salmonids, cyprinids, catfish, tilapias, European sea bass, Japanese flounder, shrimps and oysters.
