L'obésité est associée à plusieurs désordres métaboliques. L'ensemble de ces désordres sont associés à une inflammation de faible intensité dont l'origine reste peu connue. Parmi les différents facteurs environnementaux , notre laboratoire a mis en évidence différents liens existants entre l 'inflammation métabolique, le microbiote intestinal , l'obésité et le diabète de type 2. Les cellules de l'hôte entrent en communication avec différents micro­ organismes par l'intermédiaire du système immunitaire inné et plus particulièrement grâce aux « Toll-like receptors ». Ces récepteurs utilisent principalement la protéine MyD88 (Myeloid differentiation factor (88)) comme principale molécule adaptatrice impliquée dans la transduction du signal intracellulaire.Actuellement, les mécanismes moléculaires impliqués dans l'étroite relation entre le microbiote intestinal et son hôte et responsables du développement de l'obésité restent peu connus. Nous proposons que la protéine MyD88 de l'épithélium intestinal joue un rôle essentiel dans la physiopathologie de l'obésité. Pour répondre à cette hypothèse, nous avons généré un modèle de souris invalidées pour la protéine MyD88 au niveau des cellules épithéliales de l'intestin et inductible au tamoxifène (modèle VillinCreERT2-MyD88 lox/lox). Après induction de l 'invalidation , les souris ont été soumises soit à un régime contrôle soit à un régime riche en graisses et suivies pendant 7 semaines. Dans ce modèle, nous observons une diminution de minimum 75% de l 'expression de MyD88 ainsi que de la protéine, et ce spécifiquement dans l'intestin. Par ailleurs, les souris VillinCreERT2-MyD88 lox/lox résistent en partie à l'obésité induite par le régime hyperlipidique (diminution du poids et de la masse grasse d'environ 20%) par rapport aux souris sauvages. Cet effet est associé à une augmentation de la capacité de stockage du tissu adipeux (augmentation de l 'adipogenèse) ainsi qu'à une diminution de l 'inflammation du tissu adipeux (diminution de l'expression de l'ARNm de CDl l e).En conclusion, ces résultats mettent en évidence la protéine MyD88 intestinale comme un acteur clef dans le développement de l'obésité et du diabète de type 2. Ces données expérimentales supportent ainsi le concept decibler une protéine à l'interface entre le microbiote intestinal et l 'hôte pour traiter l'obésité et le diabète de type 2. Obesity is associated with a cluster of metabolic disorders. All these disorders are associated with a low grade inflammation. However, the origin of this inflammation remains unknown. Among the different environmental factors, our laboratory has contributed to the identification of various links existing between metabolic inflammation, gut microbiota, obesity and type-2 diabetes. The host cells communicate with microorganisms through the innate immune system and more particularly with Toll-like receptors. These receptors mainly use the MyD88 protein (Myeloid differentiation factor (88) as adaptor molecule involved in the intracellular signal transduction.Nowadays molecular mechanisms involved in the relationship between the gut microbiota and the host and the thereby the development of obesity are not completely understood. We hypothesize that the Myd88 protein expressed at the level of the intestinal epithelium plays are essential role in the physiopathology of the obesity.To this aim we have generated a model of mouse invalidated for the MyD88 protein in the epithelial intestinal cells and inducible with tamoxifen (model VillinCREERT2-MyD88 lox/lox). After induction of the invalidation, mice received a control diet or high-fat diet for 7 weeks. First of all, the deletion reduces MyD88 MRNA expression and protein by about 75 %. It is worth noting that we have validated that this deletion was specifically restricted to the gut. Interestingly, VillinCREERT2-MyD88 lox/lox resist partially to diet-induced obesity with a decrease body weight and fat mass by about 20% as compared with wild type mice. This was associated with restored storage capacity of adipose tissue (i.a. increase in adipogenesis and decreased adipose tissue inflammation (i.e. CD11c mRNA expression.In conclusion, our results point out MyD88 intestinal as a key player in the development of obesity and type-2 diabetes and thereby provide experimental data supporting the concept to target this protein at the interface gut microbiota and host to treat obesity and type-2 diabetes
