Abstract

Chaperokine is a term that describes the unique function of extracellular heat shock protein (eHsp) as both chaperone and cytokine. The cellular consequence of binding and signaling of eHsp is the stimulation of a potent and long lasting immune response. eHsp induces a plethora of immune responses including the release of bioactive mediators such as cytokines, chemokines, nitric oxide, apotogenic mediators, stimulation of the innate and adaptive immune response, migration and maturation of dendritic cells (DC) and the enhancement of natural killer cell-mediated cellular cytotoxicity. This edited book provides the most comprehensive review of contemporary knowledge of the chaperokine activity of heat shock proteins (HSP) in biology and medicine. Using an integrative approach to understanding the chaperokine activity of HSP, the contributors provide an overview of novel mechanisms, signal transduction pathways and a concise understanding of how the principles of the chaperokine activity of HSP has been harnessed for therapeutic gain. Key basic and clinical research laboratories from major universities, academic medical hospitals, biotechnology and pharmaceutical laboratories around the world have contributed chapters that review present cutting-edge research activities and importantly, the future direction of chaperokine research. The book is a must read for researchers, postdoctoral fellows and graduate students in the fields of Translational Medicine, Clinical Psychology, Human Physiology, Zoology, Botany, Biotechnology, Molecular Medicine, Infectious Disease, Pathology and Pharmaceutical Sciences, as well as for researchers involved in Drug Discovery.

Abstract

In this book, renowned scientists describe the role of steroid chirality and modification of lipid membrane physical properties in the modulation of G protein-coupled receptors and ion channels. The application of commonly-used technical approaches such as mass spectrometry and nucleic magnetic resonance transfer spectroscopy for studies on cholesterol distribution and alteration of lipid bilayer characteristics is also discussed. This book offers comprehensive insights into the current understanding of cholesterol-driven modulation of protein function via mechanisms that extend beyond lipid-protein direct interactions. In the first part, the chapters introduce the reader to the use of the chemical derivatives of cholesterol as a valuable laboratory tool in the studies of cholesterol-driven modulation of protein function. In the second part, examples of cholesterol-induced changes in membrane physical characteristics are presented and discussed in light of their multifaceted contribution to the effect of cholesterol on protein function. The book will be of interest to undergraduate and graduate students as well as basic science and medical researchers with a keen interest in the biophysical properties of cholesterol and physiological consequences of cholesterol presence in biological systems.

Abstract

This book presents an overview of our current understanding of the biomineralization mechanisms for shell formation in the pearl oyster Pinctada fucata, based on molecular biology, biochemistry, cell biology, structural biology and environmental biology. Pinctada fucata is the major pearl-producing shellfish in the South China Sea and is also an established model system for the research on the nacre biomineralization mechanism. Extensive studies on nacre biomineralization have provided valuable information for novel bionic material design. Discussing the isolation and gene cloning of the matrix proteins involved in the shell formation, as well as the cell signaling pathways, shell microstructures, and the environmental impacts on shell biomineralization, it is a valuable reference resource for researchers working in the field of nacre biomineralization and biomaterials.

Abstract

This book presents the latest knowledge on the roles of progranulin (PGRN) in normal physiology and pathology and explores the emerging significance of PGRN as a therapeutic target and biomarker in various CNS disorders, including frontotemporal lobe degeneration and other neurodegenerative diseases. Following initial recognition of the importance of PGRN in sexual differentiation of the developing brain and adult neurogenesis, it was subsequently discovered that PGRN acts as a chaperone of lysosomal enzymes and plays a crucial role in maintaining cellular protein homeostasis. It has also been found that sex steroids modulate the expression of PGRN and its trophic effects in the developing CNS and that PGRN directly or indirectly influences neural stem and progenitor cells. Against this background, deeper understanding of the molecular and functional properties of PGRN would provide fresh impetus for the development of mechanism-based therapeutic approaches for multiple disorders. Medications targeting the recovery of lysosomal function appear to hold particular promise in patients with neurodegenerative diseases resulting from PGRN insufficiency. In examining multiple aspects of this fascinating field, the book will be of high value for researchers and graduate students.
