Nous sommes « colonisés » par 1OO 000 milliards de bactéries résidant au niveau du tractus gastro-intestinal. Ce microbiote intestinal module différentes fonctions essentielles chez l'hôte, comme l'immunité et le métabolisme. Des données récentes obtenues au laboratoire montrent que des modifications de composition microbiote (diminution importante du taux caecal des lactobacilles) apparaissent dans un modèle murin mimant une leucémie aiguë (modèle BaF3). Ce modèle est caractérisé par la présence d'inflammation, et par un phénomène de cachexie qui est défini comme une perte de masse grasse et une atrophie musculaire.La première partie de ce mémoire fut consacrée à l'impact d'une supplémentation en deux lactobacilles, Lactobaci/lus reuteri 100-23 et gasseri 311476, dans le modèle BaF3. Les résultats obtenus confirment la restauration en ces deux lactobacilles dans le contenu caecal des souris BaF3 après supplémentation. Après analyse de paramètres de cachexie chez la souris BaF3, il en ressort que les lactobacilles n'ont pas permis de contrer la chute de masse grasse. Cependant, la supplémentation a diminué l'expression des marqueurs d'atrophie dans les muscles gastrocnémien et tibialis. La mesure de cytokines pro-inflammatoires dans le foie et le sang a mis en évidence une modulation de l'inflammation par l'administration des lactobacilles.La seconde partie de ce mémoire concerne la caractérisation d'un modèle de mélanome murin B16 et l'analyse du microbiote dans ce modèle. Les résultats montrent une chute du poids du muscle tibialis et du poids des tissus adipeux, caractéristiques en cas de cachexie. Le poids du foie est augmenté chez les souris B16, ce qui pourrait être une conséquence de l'état inflammatoire mis en évidence. Toutefois, le modèle B16 ne présente pas de modification des taux en lactobacilles, bacteroides, bifidobactéries et de bactéries totales dans le contenu caecal.En conclusion, si une dysbiose caractérise certains modèles expérimentaux de cancers systémiques et est associée notamment à l'inflammation; des études complémentaires sont nécessaires avant de proposer le microbiote comme nouvelle cible thérapeutique dans ce domaine. We are "colonized" by almost 100 trillion bacteria resident in the gastro-intestinal tract. This intestinal microbiota modulates several essential functions of the host like immunity and metabolism. Recent results obtained in Laboratory have shown perturbations of the gut microbiota (decrease of lactobacilli levels in caecal content) occurring in a model of mimicking acute leukemia (BaF3 model). This model is characterized by inflammation , decrease of lactobacilli levels in the caecal content and cachexia. Cachexia is defined as a decrease of fat mass and waste of mass muscle.The first part of this work consisted of a supplementation with two Lactobacillus, Lactobacillus reuteri 100-23 and gasseri 311476 in the BaF3 model. The results have confirmed the restoration of both Lactobacillus in the caecal content after supplementation in BaF3 mice. The analysis of cachexia parameters in the BaF3 mice showed that lactobacilli administration fails to counter the fat mass loss. However, the supplementation decreased expression of atrophy muscle markers in the tibialis and the gastrocnemius muscles. The measurement of pro-inflammatory cytokines in the liver and blood showed a modulation of the inflammation by a lactobacilli supplementation.The second part of this work was dedicated to the characterization of a murine melanoma model B16 and the analysis of the gut microbiota. The results have pointed out a decrease of the tibialis muscle weight and the fat mass,characteristic in cachexia. The liver weigh increased in B 16 mice, this may be explained by a consequence of an inflammatory condition. The B16 model did not presentany modifications in the caecal content of lactobacilli, bacteroides, bifidobacteria and total bacteria.In conclusion, if dysbiosis is characteristic for some experimental models of systemic cancers and is particularly associated with inflammation, further studies are necessary before proposing microbiota as a new therapeutic target in this area.
