Abstract

The aim of this study was to produce dry powder composed of temocillin for inhalation and optimize them for better antibiotic’s charge and enhanced aerosolization proprieties. Powders were prepared by atomization solutions of temocillin and dipalmitoylphosphatidylcholin (DPPC), main phospholipids composing the pulmonary surfactant. The DPPC’s impact on aerosolization proprieties was assessed by modifying lipids proportions in the compositions. The geometric size of the particle’s powders was assessed by laser diffraction. Tap density was calculated using a voluminometer. Aerodynamic proprieties have been studied in vitro by performing multi-sage impacters which allowed the evaluation of emitted dose and fine particle fractions. Afterwards several inhalators were tested as well modification of air flow to mimics respiratory failures. X-ray spectrometry (XPS) analysis allowed us to analyze the particle surface and to make sure of the importance of DPPC in our dry powders enhanced aerodynamic proprieties. X-ray diffraction (XRD) permitted us to know the (non-)crystalline state of the powders. Particles morphology was studied by scanning electron microscopy. Finally deposition through the pulmonary system and temocillin’s concentrations determination has been studied in vivo with mice.Samples were analyzed by HPLC after undergoing a specific extraction process to evaluate antibiotics concentration. Dans ce mémoire, nous avons comparé l'administration intraveineuse et intratrachéale de témocilline pour déterminer la meilleure voie permettant une action thérapeutique pulmonaire efficace. La déposition au sein de l'arbre respiratoire et la quantification des concentrations de témocilline a été étudiée in vivo chez des souris. Les échantillons prélevés ont été analysés par HPLC après avoir mis au point un protocole spécifique à la quantification de la témocilline. Les poudres ont été produites par atomisation-séchage d'une solution de témocilline et de dipalmitoylphosphatidylcholine (DPPC), phospholipide principal du surfactant pulmonaire . L'impact du DPPC sur les propriétés d'aérosolisation des poudres a été étudié en variant sa proportion dans les compositions. La taille géométrique des particules de poudre a été analysée par diffraction laser. La densité des poudres a été évaluée par tassement. Les propriétés aérodynamiques des poudres ont été étudiés in vitro par déposition dans un impacteur à cascades ce qui nous a permis de calculer les doses émises et les fractions des particules fines. Par la suite, ce sont différents inhalateurs qui ont été testés ainsi que des changements dans Je flux d'air pour mimer des déficiences respiratoires. Des analyses par spectrophotométrie aux rayons X (XPS) nous ont permis d'analyser la surface de nos particules et de s'assurer du rôle que joue le DPPC dans l 'amélioration des propriétés aérodynamiques de nos poudres. La diffraction aux rayons X nous a permis de savoir si les poudres produites présentaient un état cristallin. Les particules ont également été visualisées par microscopie électronique à balayage.
