Abstract

Recombinant vaccinia viruses have become important in recent years, both as protein expression vectors, and-in the age of bioterrorism-as powerful tools for developing new smallpox vaccines, diagnostics, and understanding of poxvirus pathogenesis. In Vaccinia Virus and Poxvirology: Methods and Protocols, internationally recognized experts and leaders in poxvirology describe in step-by-step detail the successful techniques they have perfected to study and work with vaccinia virus and other poxviruses. They offer cutting-edge techniques for the construction and characterization of recombinant viruses; for the study of poxvirus gene transcription and DNA replication; and for the use of the virus in immunologic and cell-cell fusion assays. Other methods cover poxvirus bioinformatics, aspects of viral pathogenesis at both a protein and an animal model level, and the study of immune responses to poxvirus-the latter critical to the important role of vaccinia virus in smallpox vaccination and its potential role as a vaccine vector directed against infectious agents and cancer. The protocols presented follow the successful Methods in Molecular Biology™ series format, each one offering step-by-step laboratory instructions, an introduction outlining the principle behind the technique, lists of equipment and reagents, and tips on troubleshooting and avoiding known pitfalls. Comprehensive and timely, Vaccinia Virus and Poxvirology: Methods and Protocols offers virologists and vaccinologists an unequaled source of proven techniques for understanding poxviruses and developing defenses against their use as bioterrorist agents.

Abstract

The recent international alarm at the possible use of smallpox as weapon of bioterror has refocused the attention of the scientific community, public health policy makers and the public on poxviruses. In contrast to the threat posed by some of these viruses, other poxviruses show exciting beneficial potential including their use as valuable tools in biomedical research and in the delivery of vaccines. This monograph provides a comprehensive review of the poxvirus family with a particular emphasis on current developments. It includes the latest insights into poxviral molecular biology, diagnosis, therapy, vaccine development and the beneficial exploitation of these viruses in biomedical research. Each chapter is written by a leader in the field. The reviews present all aspects of the field, including historical perspectives, along with summaries of the latest advances. This combination makes the book relevant for graduate students, researchers new to the field and to experienced poxvirologists.

Abstract

Orthopoxviruses Pathogenic for Humans covers those viruses capable of causing disease in man, including monkeypox, smallpox, cowpox, and vaccinia. The coverage of each virus is comprehensive, covering the biology, molecular biology, and ecology of the virus as well as the clinical and epidemiological aspects of these viruses in humans and animals. In addition, this volume highlights developments in genetic engineering that are paving the way for potential therapeutic treatments of these viruses.

Abstract

Though smallpox was eradicated from the planet two decades ago, recent terrorist acts have raised the horrific possibility that rogue states, laboratories, or terrorist groups are in possession of secret stockpiles of the virus that causes the disease, and may be preparing to unleash it on target populations.
