Presenting an area of research that intersects with and integrates diverse disciplines, including genomics, epigenetics, proteomics, and cellular biology, among others, Formalin-Fixed Paraffin-Embedded Tissues: Methods and Protocols collects contributions from expert researchers in order to provide practical guidelines to this complex study. Compiled in order to provide researchers with up-to-date methodological information pertaining to the utilization of genomic, transcriptomic, and proteomic data in diagnosis, prognosis, and tailored therapy, the ultimate aim of this volume is to decipher diseases at a molecular level. Divided into multiple convenient chapters, this detailed book covers various techniques to construct and utilize tissue arrays, it also provides detailed protocols in immunohistochemistry, immunofluorescence, fluorescent and chromogenic in situ hybridization, and ultimately introduces protocols for FFPET microdissection and nucleic acids extraction for their utilization in advanced techniques such as microarray CGH, DNA methylation and pyrosequencing. The volume also discusses FFPET research from an ethical standpoint and concludes with a chapter on novel tissue fixative. Written in the highly successful Methods in Molecular Biology™ series format, chapters contain introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible laboratory protocols, and notes on troubleshooting and avoiding known pitfalls. Authoritative and accessible, Formalin-Fixed Paraffin-Embedded Tissues: Methods and Protocols serves as a practical guide for scientists of all backgrounds and aims to convey the appropriate sense of fascination associated with this vital field of research.

The storage of biological material for regular or future use is a fundamental requirement in many biological and medical sciences. Cryopreservation and freeze-drying are the preferred techniques for achieving long-term storage, and have been applied to a diverse range of biological materials. Though the basis for many methodologies is common, laboratories frequently lack expertise with the correct storage procedures, so that many apply outdated or inappropriate protocols for storing their samples or cultures. Cryopreservation and Freeze-Drying Protocols is a compilation of the many and varied methodologies that have been developed in expert laboratories. The protocols are reproducible, robust, and in most instances have been transferred quite successfully to other laboratories. Our intended readers are those proposing to establish or improve biostorage systems in their own laboratories or units, whether concerned with culture collections, animal husbandry, aquaculture, or human fertilization programs. Because the emphasis of Cryopreservation and Freeze-Drying Protocols is on methodology, it is our intention to provide readers with the tools to make practical progress without reference to other sources. Each chapter deals with an organelle, cell, or tissue type: a short int- duction on the status of its biostorage development is followed by a detailed description of the materials required and a methodological p- tocol to be followed, with explanatory notes. This is very much a first edition; we hope and trust that future editions will contain cryopreservation and freeze-drying protocols for ceils, tissues, and organs that are at present still recalcitrant to succe- ful preservation.

Frozen sections are performed while a patient is undergoing surgery as a basis for making an immediate diagnosis that will impact treatment decisions. Frozen section diagnosis is often a highly demanding situation for the pathologist who must render a diagnosis quickly for the patient and surgeon. The Frozen Section Library series will provide concise, user-friendly, site specific handbooks that are well illustrated and highlight the pitfalls, artifacts and differential diagnosis issues that arise in the hurried frozen section scenario. The Frozen Section Library series provides concise, user-friendly, site specific handbooks that are well illustrated and highlight the pitfalls, artifacts and differential diagnosis issues that arise in the hurried frozen section scenario. Frozen Section Library: Liver, Extrahepatic Biliary Tree and Gallbladder provides a convenient reference for use during intraoperative consultation. The volume addresses commonly encountered challenges, such as the diagnosis of primary and secondary hepatic neoplasms and assessment of gross abnormalities of the gallbladder. The assessment of frozen section biopsies for liver transplantation, a challenging and increasingly encountered situation, is detailed in the form of histologic criteria that should be reported. Emphasis is placed on the differential diagnoses of gross and microscopic findings with attention to potential pitfalls and artifacts encountered on frozen section. gross and microscopic images are provided as well as user-friendly tables for quick reference.
